777-300ER也是屬于B型市場，采用目前推力最高的GE90-115BL2發動機，機身每邊設有5道登機門，其機身、機翼、尾翼、前起落架和發動機吊艙等部份都作出修改，增加了油箱容量，提高了最大起飛重量至775000磅，航程達14,685公里（7930海里）。航程增加了3550公里原因不只是單純地增加油箱容納，還采用了更具燃油效益的發動機。

777-300和777-300ER的空機重量大約是360000磅，兩者載客量和載貨量相約，但300ER型因有較高的起飛重量因而能攜帶更多燃料，使其航程增加了約34%，假如300ER型沒有增加油箱它的航程比777-300型仍多出約25%。在最大負載量的情況下，777-300型最多只能填滿60%油箱，而777-300ER則可填滿所有油箱。波音777-300ER曾在太平洋上進行ETOPS的飛行測試，最長的一次單發動機飛行達6小時29分鐘。

777-300ER計劃是由法國航空提出開發的，但法國航空并不是第一家將300ER投入營運的航空公司。首架在2002年11月14日出廠，2003年2月24日首飛，2004年3月16日取得適航證書，4月29日交付給首個客戶法國航空。日本航空為777-300ER的第二個客戶，并率先投入營運。自波音開發777-300ER以來，經歷6年時間才能夠交付給首個客戶。300ER的直接競爭對手是A340-600HGW和計劃中的A350-1000。

直至2009年9月，波音共獲得408架777-300ER訂單，已交付206架777-300ER。部份客戶以777-300ER作為過渡機種，等待波音787交付。
